Enhancing menaquinone-7 biosynthesis by adaptive evolution of Bacillus natto through chemical modulator

Abstract Menaquinone-7 (MK-7) is a kind of vitamin K2 playing an important role in the treatment and prevention cardiovascular disease, osteoporosis arterial calcification. The purpose this study to establish adaptive evolution strategy based on chemical modulator improve MK-7 biosynthesis Bacillus natto . inhibitor 5-enolpyruvylshikimate-3-phosphate synthase (EPSP synthase), glyphosate, was chosen as perform experiments. final strain ALE-25–40, which obtained after 40 cycles 25 mmol/L showed maximal titer 62 mg/L productivity 0.42 mg/(L h), representing 2.5 3 times original strain, respectively. Moreover, ALE-25–40 generated fewer spores higher NADH redox potential. Furthermore, mechanism related improved performance investigated by comparative transcriptomics analysis. Genes sporation formation were down-regulated. In addition, several genes also up-regulated. This proposed here may provide new alternative directive for industrial production K2. Graphical
